Therapeutic potential of CNP for skeletal dysplasias
نویسندگان
چکیده
Background We had revealed that C-type natriuretic peptide (CNP) is a potent stimulator of endochondral bone growth by using transgenic and knockout mice: transgenic mice with targeted overexpression of CNP in cartilage develop prominent skeletal overgrowth [1], whereas mice depleted with CNP exhibit impaired endochondral bone growth [2]. We planned to translate this effect of CNP into the therapy for skeletal dysplasias, of which the effective drug therapy is not established yet. Here we investigated the effect of CNP on the impaired bone growth of mice model of achondroplasia, the most common form of skeletal dysplasias.

Translational Research of the Activation of the C-Type Natriuretic Peptide (CNP)- Guanylyl Cyclase-B Pathway for Skeletal Dysplasia
The natriuretic peptide family consists of three endogenous ligands: atrial natriuretic peptide (ANP), brain natriuretic peptide (BNP), and C-type natriuretic peptide (CNP). They exert their biological actions through two subtypes of particulate guanylyl cyclase (GC): GC-A for ANP and BNP, and GC-B for CNP. Among the natriuretic peptide family members, ANP and BNP are cardiac hormones that are ...

Although peptides are safe and useful as therapeutics, they are often easily degraded or metabolized. Dampening the clearance system for peptide ligands is a promising strategy for increasing the efficacy of peptide therapies. Natriuretic peptide receptor B (NPR-B) and its naturally occurring ligand, C-type natriuretic peptide (CNP), are potent stimulators of endochondral bone growth, and activ...
متن کاملMeclozine Facilitates Proliferation and Differentiation of Chondrocytes by Attenuating Abnormally Activated FGFR3 Signaling in Achondroplasia
Achondroplasia (ACH) is one of the most common skeletal dysplasias with short stature caused by gain-of-function mutations in FGFR3 encoding the fibroblast growth factor receptor 3. We used the drug repositioning strategy to identify an FDA-approved drug that suppresses abnormally activated FGFR3 signaling in ACH. We found that meclozine, an anti-histamine drug that has long been used for motio...
متن کاملUnraveling the mechanism by which TRPV4 mutations cause skeletal dysplasias
Transient Receptor Potential Vanilloid 4 (TRPV4) is a mechano- and osmosensitive cation channel that is highly expressed in chondrocytes, the cells in cartilage. A large number of mutations in TRPV4 have been linked to skeletal dysplasias, and the goal of this addendum is to shed light on the mechanisms by which mutations in TRPV4 can cause skeletal dysplasias by focusing on 3 recent publicatio...
متن کاملRole of the natriuretic peptide system in normal growth and growth disorders.
The C-type natriuretic peptide (CNP) and its receptor (NPR-B) are recognized as important regulators of longitudinal growth. Animal models involving CNP or NPR-B genes (Nppc or Npr2) support the fundamental role of CNP/NPR-B for endochondral ossification. Studies with these animals allow the development of potential drug therapies for dwarfism. Polymorphisms in two genes related to the CNP path...
